چکیده

Problem statement: The aim of study was to investigate the effect of Salivary Gland Extract (SGE) of Lucillia sericata maggots on Human Neonatal Fibroblasts (B-HNF-1) proliferation and migration within collagen/hyaluronan membrane. Chronic, non-healing wounds are major health care problem worldwide. Nowadays, Maggot Debridement Therapy (MDT) has attracted much attention due to its successfull application in a treatment of chronic wounds where conventional treatment has failed. Maggot Excretion/Secretion (ES) contains specific proteinases which are positivelly involved in the remodeling of Extracelallar Matrix (ECM) components. Approach: Immunohistochemical methods using primary monoclonal antibody against marker vimentin was employed for the visualization of fibroblasts cultured within collagen hyaluronan membrane. Results: We found that maggot SGE at concentration of 2.4 and 4.8 gL mL significantly induced the migration of fibroblasts into the membrane after 5 and 10 days of cultivation. On the other hand, SGE at concentration of 7.2 gL mL reduced the migration of fibroblasts. Conclusion: The obtained results suggest that medicinal maggots secretion may speed up the wound healing process through the stimulation of fibroblast migration. However, further detailed research is needed to fully elucidate the substance (s) responsible for fibrloblast stimulation and migration.

The Role of Human Adult Peripheral and Umbilical Cord Blood Platelet-Rich Plasma on Proliferation and Migration of Human Skin Fibroblasts

BACKGROUND Wound healing is a complex and dynamic process following damage in tissue structures. Due to extensive skin damage caused by burn injuries, this study determined the role of human adult peripheral and umbilical cord blood platelet-rich plasma on proliferation and migration in human skin fibroblasts. METHODS Platelet-rich plasma (5, 10, 15, 20 and 50% PRP) from human umbilica...

Characterisation of the Small RNAs in the Biomedically Important Green-Bottle Blowfly Lucilia sericata

BACKGROUND The green bottle fly maggot, Lucilia sericata, is a species with importance in medicine, agriculture and forensics. Improved understanding of this species' biology is of great potential benefit to many research communities. MicroRNAs (miRNA) are a short non-protein coding regulatory RNA, which directly regulate a host of protein coding genes at the translational level. They have been...

EFFECTS OF DIFFERENT MEDIATORS ON FIBROBLAST PROLIFERATION

In this study, we have investigated the proliferative response of human dermal fibroblasts isolated from normal and psoriatic individuals to different cytokines. We found that IL-1, TNF (α) and EGF are stimulatory whereas IFN « and 1 both showed inhibitory activity on fibroblast proliferation. IL-6 a more recently described cytokine, showed no detectable effects on cell proliferation. Inve...

Development of an Allogeneic Cultured Dermal Substitute Using a Standard Human Fibroblast Bank

Background: Fibroblasts are mesenchymal cells that can be readily cultured in the laboratory and play a significant role in epithelialmesenchymal interactions, secreting various growth factors and cytokines that have a direct effect on epidermal proliferation, differentiation and formation of extracellular matrix. They have been incorporated into various tissue-engineered and used for a variety...
